Water accessibility is the exposure of a biological structure to nearby water molecules. The interaction of water with proteins is essential for their structure and function, affecting protein folding and dynamics. Hence, applicability of the method is tested on a well-documented and simple model GB1 protein, which is a β1 immunoglobulin G-binding protein G derived from the streptococcus bacteria. The method employs fast magic-angle spinning (MAS) of the sample at 54.7° to average out anisotropic interactions and reduce line broadening. At 60 kHz, spin diffusion is partially suppressed so that a combination of spin diffusion and nuclear Overhauser effect (NOE) is responsible for polarization transfer. This study investigated the roles of padR and cwp9, both upregulated during early colonisation. Overexpression of padR resulted in a significantly slower doubling time and reduced biofilm formation, suggesting that PadR functions as a transcriptional regulator controlling genes linked to colonisation and bacterial fitness. In contrast, cwp9, predicted to be a cell wall-associated hydrolase, conferred only an initial growth advantage without affecting overall growth or biofilm formation. Understanding the functions of these proteins may inform the development of novel therapeutic strategies to prevent C. difficile colonisation and infection. Alongside this trend, anti-populism has emerged as a strong counterforce, critiquing the rise of populism and populist ideas. Scholars have shown that anti-populism, empowered through dominant public and media discourses on populism, has been a powerful lens through which to define the concept of populism itself. The latter has become increasingly misunderstood as a danger to democracy, while there in fact exist a variety of populisms, left- and right-wing, democratic and anti-democratic. This research project will therefore explore the difficulties associated with defining both populism and anti-populism as concepts and investigate the democratic potential of left-wing populist movements that have developed in response to democratic crises. Finally, I will explore how the media engages with populist ideas, figures and parties. This will include a focus on how the written media practises agenda-setting and deflection when discussing populism, as well as the role of populist hype in the euphemisation, trivialisation and amplification of populism through The Guardian’s articles. Parameter-efficient fine-tuning (PEFT) methods like Low-Rank Adaptation (LoRA) address this by updating only small low-rank matrices while keeping pretrained weights frozen. This project explores LoRA and its variants—QLoRA, AdaLoRA, and LoRA-One—through theoretical analysis and experiments on the GLUE benchmark using T5-base. Results show that LoRA achieves near full fine-tuning performance with under 1% of trainable parameters. The study highlights LoRA’s efficiency–expressiveness trade-off and contributes to developing more scalable, adaptive fine-tuning strategies for large models.
